Palmitoyl tripeptide-5 (Palmitoyl-Lysyl-Valyl-Lysine, Pal-Lys-Val-Lys-OH) is also called "tripeptide" or "collagen peptideCOLL-O2 ", the most widely used cosmetic peptide. Palmitoyl tripeptide-5 belongs to the signal peptide class and can promote the synthesis of collagen (I type, II types, and IV type) and the production of extracellular matrix protein by increasing the activity of stromal cells to make the skin looks young and elastic. Collagen can be added to cosmetics to help skin supplement collagen, improve facial elasticity and remarkably reduce facial wrinkles. Meanwhile, the skin care product has the characteristics of enhancing cell activity, increasing the skin’s water content, enhancing the skin luster, and improving the skin color.
In the prior art, more research reports about applying palmitoyl tripeptide-5 exist. As a result of research on the whitening efficacy and action mechanism of palmitoyl tripeptide-5 in the literature [ daily chemical industry, 2018,48(3),166-171], the palmitoyl tripeptide-5 can inhibit the generation of melanin in melanocytes by inhibiting tyrosinase activity and reducing mRNA levels of microphthalmia-associated transcription factor (MITF) and tyrosinase and can be applied to the field of cosmetics as a whitening additive. Chinese patent publication No. CN108619047A discloses an anti-aging cosmetic formulation and a preparation method, wherein one of the anti-aging components is palmitoyl tripeptide 5. Chinese patent with publication No. CN 107669564A discloses a skin care composition with repairing and regenerating, moisturizing, and delaying senility, wherein palmitoyl tripeptide-5 is one of the main components. Many reports of related research need to be listed here.
However, the literature on preparing palmitoyl tripeptide-5 in the prior art is scarce, and the practice of palmitoyl tripeptide-5 by a solid phase synthesis method is mentioned in U.S. Pat. No. 5, 2007099842, 1, but various protecting groups and deprotecting reagents are required, and the final yield is only 19%. Besides the method, this is the only method for preparing the palmitoyl tripeptide-5 reported at present. (CN201910789414.XA, 2020)
Bio scientific research. It is not obtained from animals. Palmitoyl tripeptide-5 is a laboratory peptide comprising three amino acids: lysine, valine, and lysyl.
Its three amino acids can penetrate deep into the epidermis and even the dermis. They work from the inside out to increase collagen production, allowing you to achieve better results faster than animal-derived collagen.
Palmitoyl tripeptide-5 acts on specific skin receptors and stimulates collagen production in our skin cells. It firms our skin and reduces the appearance of wrinkles and fine lines. It makes your skin look healthier and fresh.
Palmitoyl Tripeptide-5 also interact with hyaluronic acid and elastin production system. The combined effect is increased water content, reduced skin wrinkling, and enhanced skin firmness.
Additionally, it supports the healthy functioning of the skin cells and helps remove toxins.

SynColl Research Findings
Although a relative discovery, Palmitoyl Tripeptide-5 is used in cosmetic formulations that aim to provide anti-aging benefits such as reducing fine lines and achieving smooth skin, especially for sensitive skin.
In-vitro studies have also shown that Palmitoyl Tripeptide-5 can prevent collagen breakdown by avoiding the degradation pathway of Type 1 and Type 3 collagen in the skin. The studies also indicate that this synthetic peptide can boost collagen synthesis.
New products are also being developed using Palmitoyl Tripeptide-5 and other raw ingredients to help reduce redness in Type 1 rosacea. The active complex inhibits stimulating inflammatory proteins known as cytokines in the human body.
Benefits of Syn-coll for Skin: (DSM, SYN®-COLL)
Visibly reduces the appearance of wrinkles in age-prone zones: -
SYN-Coll: Power Anti-aging with a Collagen-boosting Peptide
Skin aging, in particular when induced by chronic ultraviolet (UV) exposure, leads to degenerative changes in the skin. Ultraviolet light also increases metalloproteinase activity. This leads to collagen breakdown at a higher rate than just normal aging. The consequences are manifold, such as increased skin fragility, leathery skin appearance, and the formation of superficial fine wrinkles. If not taken care of, these fine lines become visible to others.
Consequently, anti-wrinkle products are the first choice if consumers want to reduce the appearance of wrinkles or to stave off the first signs of aging.
SYN-Coll, a small molecular synthetic peptide (see Figure 1), has been designed to prevent the formation of wrinkles and reduce their appearance. Peptides comprise amino acids, the major components of all living cells, including skin cells. Peptides are beneficial ingredients in cosmetic formulas; they help revitalize the skin and counter the effects of UV-induced premature aging.
Collagen represents the main component of the dermal connective tissue's Extracellular Matrix (ECM). Thrombospondin I (TSP) is a multifunctional protein that activates the latent but biologically inactive form of the Tissue Growth Factor (TGF-β), the crucial element in synthesizing collagen. Like TSP, our peptide molecule binds to latent TGF-β and initiates its activation to fully functional TGF-β (demonstration in vitro), as shown in Figure 2.
This innovative mechanism made our peptide a unique and specific TGF-β booster. It comprises the increase of collagen and simultaneously protects collagen from degradation through MMP inhibition (demonstration in vitro). Comprehensive clinical tests show strong results on the appearance of all types of age-related wrinkles (see Figure 3) and have confirmed their safety for cosmetic use.
Significantly reverses visible signs of photo-aging damage: -
It helps to slow down the skin aging process and reverses photo-aging damage. It offers a lifted, sculpted look and refines a pore appearance for smoother, younger-looking skin. Moreover, it offers improved skin firmness, elasticity, texture, and smoothness.
Rejuvenates and smoothens the skin: -
Delivers voluminous moisture deep into the skin, encouraging depleted cells to regain their fullness and improving the skin's overall appearance.
It is also thought that palmitoyl tripeptide-5 may interact with hyaluronic acid and elastin production systems. The combined effect of these characteristics is increased water content, improved appearance of fine lines and wrinkles, and a thickening of the skin. Additionally, studies have suggested that it helps support the healthy functioning of the cells, improving toxin removal.
Refines pore appearance & improves skin texture: -
Pores play an essential role in the skin's health. They allow your skin to breathe, eliminate toxins, and cool off. As such, they keep the skin healthy. Pores are small openings of hair follicles on your skin. However, enlarged pores can be problematic. They can lead to bacteria build-up and clogging. Unclean and unhealthy pores can cause various skin conditions, including acne.
With aging, your skin’s ability to produce essential skin proteins called elastin and collagen reduces. This leads to the loss of skin elasticity, ultimately resulting in bigger pores, wrinkles, and saggy skin. Your pores contain sebaceous glands that produce oily, waxy sebum.
Ageless hydrates and replenishes your skin's suppleness, returning it to its natural plump fullness.
It targets the Nasolabial fold lines (the vertical parenthesis around the mouth), forehead lines, under-eye hollowness, and crow's feet by smoothing and filling in for a lipo-filling like-effect plus visibly improves the look of fine lines and wrinkles.
Improves skin firmness and elasticity: -
Over time, your skin's elasticity will improve, leaving your skin looking and feeling firmer and lifted. In addition of it, syn-coll acts as a wrinkle filler to help smooth out pockets that have developed over time. As a result, your complexion will appear refreshed and significantly younger-looking.

Uses of Syn-coll:
How Peptides in Your Skin Care Work:
If peptides are already present within the body, why do we need more of them? The answer, according to experts, is aging. “We lose 1% of our remaining collagen per year after age 30,” says Dr. Robinson. “Our skin’s natural communication channels also slow down over time.”
But simply slathering peptides onto your skin doesn’t necessarily give you more of them. According to Dr. Bowe, their work is a little more complex: “Your body sees these peptides as ‘signals’ that you need to heal, which tells your body to produce more collagen where you need it most,” she explains. “If you are healing from an injury in your ankle, those peptides will signal your body to focus its repair efforts in that joint. If your skin is aging prematurely, those peptides might signal your skin to boost its production of collagen and even hyaluronic acid, plumping up your skin and restoring a healthy skin barrier. When you introduce exogenous peptides onto the skin as a moisturizer or serum, it tricks the skin into thinking there’s been an injury or wound and stimulates our collagen-boosting processes.”
Side effects and risks:
The Cosmetic Ingredient Review Expert Panel, a group that reviews the safety of skincare and cosmetic ingredients, has reviewed the available data on palmitoyl oligopeptides and has determined that they are safe for use. There are no reported sensitivity, toxicity, or hormone disruption issues. However, palmitoyl tripeptide-5 is a relatively new ingredient. While there is a respectable amount of research on its effects, further information will be reviewed by the Cosmetic Ingredient Review Expert Panel. As they mentioned in their report, given the similarity in structure to other peptides in the body, they have the potential to interact with different pathways in the body.
Palmitoyl Tripeptide-5 is a new ingredient, and only some researchers are on it. But according to a skincare specialist, Palmitoyl Tripeptide-5 is safe to use on any skin. It can’t react to your skin. There is no reported toxicity, allergy, sensitivity, and hormone disruption. If you have sensitive skin, you may respond to anti-aging creams. Your everyday skin reactions are redness, itching, and stinging at the application site. If you suffer from any of these side effects, you should rush to your dermatologist to see whether palmitoyl tripeptide is the main culprit and if you can apply an alternate.
The only disadvantage:
The only disadvantage of this synthetic product is that it needs to be better researched. Many researchers are ongoing to prove its authenticity. (EBLOUIR GROUP, 2021)
